The spring barley (Hordeum vulgare) was modified for altered stomatal function and sensitivity to ozone. HT1 encodes HIGH LEAF TEMPERATURE 1, a Raf-like protein kinase associated with guard-cell signalling and the regulation of stomatal movements in response to changes in carbon dioxide concentration. Modification of this locus may alter CO₂-dependent stomatal regulation, stomatal conductance and gas exchange, thereby affecting ozone uptake and sensitivity, since ozone enters leaves primarily through open stomata. The line is intended for research on the relationship between carbon-dioxide sensing, stomatal regulation, ozone response and barley performance.

Information on the modified DNA sequence
The hv_ht1 line was developed through CRISPR/Cas-mediated targeted mutagenesis of a barley locus related to HIGH LEAF TEMPERATURE 1 (HT1), using the Sm13-cas9-ec64420_Barley vector. HT1 proteins are Raf-like serine/threonine protein kinases associated with guard-cell signalling and the regulation of stomatal movements in response to changes in carbon dioxide concentration. Their molecular function has been characterized primarily in Arabidopsis thaliana, where mutations in HT1 disrupt normal CO₂-dependent stomatal responses.

The plants selected for the field trial are homozygous for the targeted modification. Their progeny does not contain transgenic sequences originating from the CRISPR/Cas expression vector used to induce the mutation.

The modified locus can be characterized by PCR amplification of the target region followed by sequence analysis.
